Inter have been chasing the signature of Mancini since the arrival of Mourinho earlier this summer.
Mancini has inked a four-year deal with Inter after a fee in the region of €12.5million (£10million) was agreed with Roma.
The Brazilian was unveiled as their first summer signing on Tuesday afternoon.
Mancini told a news conference: "I thank Roma for everything they have done for me, for how they have treated me and for the way in which they have behaved in the negotiations with Inter."
Meanwhile, young midfielder Francesco Bolzoni has signed a new deal with the Nerazzurri
The 19-year-old has extended his contract until 2012 with the Italian champions after progressing through the youth system at the Giuseppe Meazza.
